Center:When finished with the setup, push Start experiment (in the experiments menu) to startacquire the Z-stack. Go to Live mode, use thefocus knob to find the centerof the sample and pushCenter to select the centerposition of the sample2. Select Slice and give the slicenumber for the total rangeof the Z-stack. This will bedivided above and below thecenter position. SelectInterval and type in thedesired interval betweentwo stack. If unsure, pushOptimal button to not loseany information between 2stacks.3. Optimize the pinhole for allthe channels. Note that thisonly works for LSM mode.4. Set up gradient intensity fordeeper samples (if needed)
