Using a calibrated positive displacement micropipette,dispense 400 μL Hemoglobin Denaturant into amicrocentrifuge tube.NOTE: Dispense Hemoglobin Denaturant directlyto the bottom of the tube; do not allow thedenaturant to drip down the inside of the tube.2. Using another calibrated positive displacementmicropipette, withdraw exactly 10 μL of the well-mixedwhole blood patient specimen or control.3. Insert the pipette into the microcentrifuge tubecontaining the Hemoglobin Denaturant, allowing the tipof the pipette to barely make contact with the surface ofthe denaturant. Dispense the specimen (1:41 dilution).4. Withdraw and redispense the mixture two times, keepingthe tip of the pipette in constant contact with fluid in thetube.5. Mix by gently vortexing at medium speed 5 to 10 seconds.Avoid foaming.6. Prior to testing, allow the hemolysate to incubate for atleast 5 minutes at 15 to 30°C.7. Transfer hemolysate to a sample cup and place on theinstrument.
