27SECTION 6 – QUANTITATIONThe quantitation measurement mode enables sample concentrations to be calculated using astandard curve. In this mode a number of standard solutions covering a range of knownconcentrations are measured. The absorbance or % transmittance of these solutions is plotted tocreate a standard curve. Once the standard curve has been created a sample of unknownconcentration can be measured and the concentration calculated using the standard curve.6.1 QUANTITATION MODE SCREENThe quantitation measurement mode enablesmeasurement parameters to be changed. The twooptions at the bottom of the screen allow the user toselect the unit of measurement and edit the calibrationstandard settings. These options are selected with thecorresponding function buttons.6.2 WAVELENGTH SELECTIONThe wavelength can be adjusted by pressing the GOTO key on the instrument keypad. The user canselect from the following options:1. Single WL2. Isobestic where Abs = Abs1 – Abs23. 3 PointsThe required wavelength values can then be entered using the numeric keypad. Press the Enter keyto confirm.6.3 METHOD SETUPIn this measurement mode the method setup parameters are accessed by pressing the function keysbelow the two options on the quantitation menu screen.m x Absλ1 + n x Absλ3where: Abs = Absλ2 - m + nm = Absλ3 - Absλ2 and n = Absλ2 - Absλ1
