CHAPTER 1 - SYSTEM OPERATIONZEISS Left Tool Area and Hardware Control Tools LSM 880104 000000-2071-464 10/2014 V_015.2.8.5 HDR(High Dynamic Range)-ImagingHigh dynamic range (HDR) creates an image with adepth of 32-bit therefore covering a far broaderdynamic range than conventional images (8 to 16bit). HDR-imaging is available in the Show allmode.HDR creates the typical HDR effect of boostingweak structures without saturating bright areas inthe image thereby enabling an optimalrepresentation of the morphology of weak andbright objects within the same image. Please see the description of the processing toolHDR-Imaging (section Processing – HDR-imaging) for details on the properties andprocessing options of HDR files. The HDR function is located under the fieldAveraging in the Show all mode of theAcquisition Mode tool (Fig. 130). HDR isactivated through ticking the HDR checkbox.HDR is exclusively available to LSM imaging and isnot selectable when using other modes of imagerecording (e.g. WF, SIM and Laser WF imaging onELYRA systems). Two methods are available (Fig. 131):− Sampling (HDR)− Illumination (ROI HDR)The HDR method used for image capture is also shown in the information of the recorded images. The Sampling method is a means to improve the image display as it equalizes the differences betweenthe very high and the very low intensities in the image. However, it is not suitable for quantitative analysisas the grey level steps become non-linear.Fig. 130 Acquisition Mode tool, HDR panelFig. 131 HDR modes
