CHAPTER 1 - SYSTEM OPERATIONZEISS Left Tool Area and Hardware Control Tools LSM 880132 000000-2071-464 10/2014 V_015.2.13 Acquisition Parameter – Shuttle and FindShuttle and Find allows calibration and imaging of sample positions in laser scanning microscopes (LSM)for reproducible repositioning after transfer to scanning electron microscopes (SEM).For Shuttle and Find different types of sample holders are pre-designed. In addition the user can defineother correlative sample holders.The correlative sample holders have three fiducial markers enabling a three point calibration.The calibration can be done in transmitted light or reflected light.A motorized microscope stage is mandatory.The workflow is unidirectional from LSM/SR to SEM only.Different types of sample holders are available(Fig. 174), e.g.:− Life Science cover glass 22x22− Life Science for TEM Grids− MAT Universal A− etc.The Life Science cover glass 22x22 or thesample holder for TEM Grids should not beused together with Axio Imager andAxio Examiner equipped with a leveladjustable sample holder. During thecalibration procedure the objective orneighbouring objectives will collide with thelevel adjustable holder.Creating of user-defined correlative holdersIn the calibration dialog window new correlativeholder files can be defined (Fig. 175).• Click on the + button in the Select Templatedialog.• Type in a name for the new holder / samplecarrier. An image of the new holder can beloaded.In the holder file the distances (in millimeters)between the first and the second marker andbetween the second and third marker have to beinserted.The distances can be determined using the Positions button in the Dimensions control view and thefunctionality for Marks in the Stage tool in the Left Tool Area.• Activate the Positions button and click on the first correlative marker.• Determine the coordinates by clicking on Mark in the Stage tool.• Go on with this procedure for marker two and three.
