The tool has three tabs:− Data,− Measure,− Auto Save.5.5.6.1 FCS Options – DataSaving Raw Data During MeasurementIf the Save raw data during measurementcheck box is activated (Fig. 582), raw data will besaved temporarily in a specified directory on disc(Fig. 583). Each single measurement will be savedin a single raw data file. The name of the raw datafile is an identifier to the .fcs measurement itbelongs and the channel. This option is used whenaccess to the raw data is required to analyze thedata in a different way in respect to correlationand PCH settings or within other programs.If the check-box is de-activated, no raw datawill be saved.Temporary raw data will be deleted from thespecified file upon closing the Image FCSdiagram. You will be prompted in the Saveraw data dialog box, if you want to save theraw data. Press Yes to save the raw datapermanently along with the .fcs file in aspecified directory, which can be defined inthe Save dialog box. Press No to discard theraw data. Note that raw data are savedalong with the .fcs measurement file in thesame folder. They are attached to the .fcs filevia a relative file path. If you save the samemeasurement again without the option SaveRaw Data, than the raw data files will bereassigned an absolute file path and theconnection to the .fcs file is lost.Please note, if the Auto safe checkbox is ticked in the Auto Save submenu, temporary raw datawill be stored permanently and have to be deleted manually. Care has to be taken to avoid discspace limitations, since raw data files can be huge.Each single measurement is stored in one raw data file.
