The size of thefinal tiled image depends on the settings for theimage format set in the Acquisition Mode tooland the number of tiles.The application of the Tile Scan function requiresan objective with a minimum magnification factorof 2.5x.The number of Tiles can be defined in the TilesHorizontal and Vertical Input boxes. A maximumof 100 tiles can be set. The size of the final TileScan is displayed in Pixels and as a Size in μm.According to image format the number of Pixelschanges. According to the objective used forimaging the Size of the Tile Scan changes.The Tile Scan grid is displayed in the grid displayarea. It shows the number and location of thesingle tiles and also indicates any overlap.The Tile scan is created around the absolutestarting position of the stage. In case the stagedoes not move exactly horizontal to the scan fielda Rotation value can be set in the Rotation inputbox to correct for this (available only in Show allmode). This can either be calibrated beforehandusing a grid slide or any other specimen whichdisplays clearly visible structures in horizontaldirection.Acquire smaller tile scans to see the effect of aspecific value for Rotation until you find the rightone. This value should be taken for the Rotation.Alternatively, use the Tile Scan Rotation Macrowhich provides an automatic Rotation calibrationfunction. Take over this value to the main softwarebefore acquiring the tile scan.When Bi-directional is active (available only inShow all mode) the image acquisition isperformed also on the horizontal backwardsmovement of the stage.If the system is equipped with the software option TILES, the Overlap input box is available (Fig. 205).Define the overlap of the tiles as the % of the tile size. The extent of overlap will also be displayed in thegrid display area.
