CHAPTER 1 - SYSTEM OPERATIONZEISS Left Tool Area and Hardware Control Tools LSM 880162 000000-2071-464 10/2014 V_015.2.20 Multidimensional Acquisition –PositionsWhen the Positions tool tab is expanded thePositions tool becomes available (Fig. 234).This function comprises two main applications.First, it is used to take images of a specimen atfreely defined positions (Positions List mode).Second, it is used to image specimens, typicallycells, which are grown in sample carriers like plateswith a set number of wells (Sample Carriermode).5.2.20.1 Position ListPress the Position List button to obtain thePosition List control panel (Fig. 235)Clicking the Add button adds the current positionand its coordinates to the list of positions.Displayed are the number of the positions, their x(μm), y (μm) and z (μm) coordinates and alsotheir offset values (μm). See the description ofSetting individual offset values for positionsin Position List mode (below) and also the tooldescription of Focus Devices and Strategy fordetails on offset values (section AcquisitionParameter – Focus Devices and Strategy).Highlighted positions can be removed by pressingthe Remove button. Alternatively the whole listcan be deleted by pressing the Remove Allbutton.It is also possible to move the stage to thehighlighted position by clicking the button Move to. The order of positions in the list can be modified bypressing the buttons Up or Down.You can save and load pre-saved positions by pressing the Save… and Load… buttons, respectively,which will open Windows Explorer.The list of positions is consistent with the positions defined in the Stage tool.When the list of positions is deleted in the Position list, it is also deleted in the Stage tool. Thisis not the case the other way round.It is recommended to save the position list as relative positions. The format "Positons list files(relative)" should be selected in the Windows Explorer. This format allows to reuse the positionsfor other experiments later on. Just make sure that the reference position for the list (forexample the position 0) matches with the actual zero position in your sample.
