Use the MBS 488/561 as the maindichroic beam splitter. We presume that nopredefined configuration is available for thisimaging task.The description applies to the use of anymicroscope.Data acquisitions is impaired whenkeeping the computer busy with othertasks then acquisition like processing dataor using other software in the mean time.This might slow down image acquisitionand can even interfere with the timing ofthe acquisition.5.2.23.1 Requirements• Position the specimen under the objective andfocus onto the desired imaging plane.• Activate Show all Tools and open theImaging Setup tool. Chose Channel mode forthe image acquisition (Fig. 249).• Switch on the Laser 561 nm.• Click on the laser icon, select the 561 line, andset the transmission slider to about 3 % (thislaser has an output power of 10 mW, otherlasers have up to 100 mW depending on thetype of laser and the imaging device used; becareful when setting the power for imagingwith other lasers for imaging other dyes).
