CHAPTER 1 - SYSTEM OPERATIONLSM 880 Left Tool Area and Hardware Control Tools ZEISS10/2014 V_01 000000-2071-464 3615.4.19 Processing – Lightsheet ProcessingLicensing Instrumentstandard optional LSM ELYRA Lightsheet Z.1See full description of tool set for details. The Lightsheet Processing module allows one to process images that were acquired with theLightsheet Z.1 using Dual Side illumination and/or with the Multiview tool tab.Files need to be saved prior to any Lightsheet Processing task. If this is not the case the software will askto save the data before proceeding. All images and registration files resulting from Lightsheet Processing can be found in a Resultfolder that is created in the folder of the original file, that was used for processing.5.4.19.1 Dual Side FusionThis processing task can be used for the followingdatasets acquired with Dual Side illumination:− Image (Snap function)− Time Series− Z-Stacks− Combinations of Time Series and Z-Stacks− Multiview experiments as well incombination with Time SeriesWhen a dataset has been acquired using DualSide illumination, the dataset will have theillumination dimension saved in its metadata andthe Dimension slider for Illumination is visible inthe Image Control area. Both illumination sidesare handled as single frames within the image.Dual Side Fusion provides three main options formerging both images from the differentillumination sides into a single frame.Choose the dataset you wish to perform the taskby mark either option (Fig. 515).Discrete Fourier Transform Fusion:The fusion is performed based upon the published method ("Multiple imaging axis microscopy improvesresolution for thick-sample applications", Jim Swoger, Jan Huisken, and Ernst H. K. Stelzer, OPTICSLETTERS / Vol. 28, No. 18 / September 15, 2003).Fine structures and details are more visible after the fusion which might be worsened when using MeanFusion. However, this process can create noticeable artifacts in the resulting image.Fig. 515 Processing – Lightsheet Processing.Dual Side Fusion
