OPERATIONQuick start LSM 5104-18 B 40-051 e 07/98(17) To further improve image quality you can slow the scan rate allowing more photons to integrateon the detector, or apply image averaging to remove random noise, or a combination of both. These adjustments are made by clicking on the button on the Scan Control menu. Setthe Scan Speed in the Speed area and Number in the Depth, Scan Direction & Scan Averagearea accordingly by observing your image. The setting average of 8 (Number 8) should improvesignal/noise dramatically, however, the image acquisition rate will be slower.(18) If your specimen is sensitive to photobleaching, you can attenuate the laser illumination by clickingon the button from the Scan Control menu. At the bottom of the menu you can setthe percentage of laser power (Excitation %) for each excitation wavelength. You will probablyhave to inrease the Detector Gain if you decrease the laser power. This setting controls thetransmission degree of the AOTF.(19) To save your image click on the or button on the right side of the image.Fig. 4-18
