CHAPTER 4 - SYSTEM OPERATIONCarl Zeiss Left Tool Area and Hardware Control Tools Lightsheet Z.1128 000000-1790-528 02/2013If the stack is large, available memory might belimited. In this case a Warning is issued, recommending the usage of the 3D large mode(Fig. 197). In this case, the stack is subdivided intoindividual smaller stacks, which are individuallyprocessed. The result images of the individualstacks are than automatically combined into oneoutput image. If one selects a region of interest (ROI) in theimage, than that ROI will be used for imageprocessing. This also can help in reducing memoryproblems when processing huge stacks. In the Processing drop down menu one can selectbetween Automatic and Manual processing(Fig. 198). If Automatic is selected, pre-set filterand parameter values will be used.If Manual is selected these filters and parametersbecome accessible to be changed and the Manualselection field becomes accessible (Fig. 199).The Automatic mode is recommended forimages with high modulation contrast. Incase the modulation contrast is low onemight benefit from changing the pre-setvalues.In the Channel drop down menu (Fig. 200) All orsingle channels can be selected from a multi-channel image that should be processed. TheChannel drop down menu is not displayed if theloaded image has only one channel.Due to the limited number of rotations morefrequencies and hence higher resolutioninformation can be obtained exactly at the rotationangles. The directions between the rotationalangles contain lesser frequencies. All availablefrequency information will be used if theMax.Isotrop checkbox is not ticked. This gives riseto the flower like appearance of the Fouriertransform, which extends wider at the rotationalangles.Fig. 197 Warning for memory shortageFig. 198 Processing drop down boxFig. 199 The Manual selection fieldFig. 200 Channel drop down box
