CHAPTER 1 - HARDWARECarl Zeiss User Interfaces Lightsheet Z.132 000000-1790-528 02/20133.5 Assembly of the Sample HolderDepending on the size of the sample there are two different types of sample holders available: sampleholder for capillaries (inner diameter of capillary size 1 / ~0.68 mm, size 2 / ~1 mm, size 3/ ~1.5 mm,size 4 / ~2.15 mm) and syringes (1 ml). Always use the minimal cylinder diameter necessary for yourspecimen size to avoid excessive amounts of embedding medium.3.5.1 Assembly of the Sample Holder for Capillary1. Select the corresponding colored sleeves (Fig. 26/3) to match the color of the capillary of choice(Fig. 26/6).2. The capillary should hold the sample and the appropriate plunger (Fig. 26/5), see as well theCHAPTER 2 SAMPLE PREPARATION in this manual. The plungers that fit into capillary size 2 - 4have to be used with corresponding Teflon tips that are already assembled onto the plungers. Notethat in addition 10x Teflon tips of each as well as matching Teflon tip tools are provided in theChamber & Sample Holder Starter Kit Lightsheet Z.1 (Tab. 1).
