CHAPTER 4 - SYSTEM OPERATIONLightsheet Z.1 Left Tool Area and Hardware Control Tools Carl Zeiss02/2013 000000-1790-528 1753.4.5.3 Adjustment – Manual DetectorAlignmentThis adjustment is necessary for one or twodetection modules when you recognize a pixel shiftbetween channels or when the zoom is notcentered.The Automatic Detector Alignmentshould always be performed first since itmight already solve the problems.Furthermore, it defines the settings for thecombination Channel 1 (Cam1) and position1 of the Emission Selection Filter (see section3.2.5.7 Emission Selection FilterSelection), which should be used as areference during the Manual DetectorAlignment.The Zoom in the Acquisition tool windowis set to 1 while performing the ManualDetector Alignment.No sample should be present when performing thistask. When pressing the Manual DetectorAlignment button, a window will open (Fig. 272).A grating is automatically inserted into the lightpath and a live image of it is shown in a newimage container within the ZEN software. The SetExposure button is activated automatically toadjust brightness. All components of the light pathcan be controlled in the Manual DetectorAlignment window. The light source is the LEDwhite light source to ensure the visibility of thegrating independent of the filters in the light path.All other controls for adjusting imaging settingsshould be controlled in the tool windows of themain software.Fig. 272 Manual Detector Alignment window
